For those traveling by air, Sag Harbor is approximately a two hour drive from JFK and LGA airports. Islip (ISP) is a closer option with limited flights. Avoid Newark (EWR) as it is significantly further.

Car rentals are the most economic and convenient option. Ubers from the airports run approximately $350-400 each way. Once in the Hamptons, Ubers are aplenty.
